Homo sapiens is unique among primates in its habitual, efficient bipedal locomotion. While many theories emphasize the adaptive advantages of upright walking — such as thermoregulation, provisioning, or predator defense — fewer address its genetic and developmental origin. This paper proposes that the fusion of two ancestral ape chromosomes into modern human Chromosome 2 acted as a developmental trigger for bipedality. This rare genomic restructuring, evidenced by telomeric remnants and a vestigial centromere, re-architected chromatin topology near the HOXD cluster (2q31). By resetting nuclear positioning, folding dynamics, and CTCF boundary activity, the fusion indirectly biased HOXD enhancer timing, prematurely truncating iliac growth. The result was a transformed pelvis — from a planar, ape-like structure to a shortened, cylindrical configuration — that reoriented gluteal musculature for upright posture. Fossil evidence, particularly the mosaic anatomy of Australopithecus afarensis ("Lucy"), aligns with this developmental shift: an arboreal upper body paired with a pelvis already constrained for bipedality. In this framework, bipedality originated as a developmental byproduct of chromosomal fusion, later refined by natural selection acting on distributed regulatory networks (SOX9, RUNX2, FOXP1/2, PTH1R, ZNF521). This two-stage process distinguishes between the origin of upright locomotion — a genomic accident — and its refinement through adaptive pressures.
